搖床

搖(yao)(yao)床(chuang)又稱搖(yao)(yao)瓶機,它是培養(yang)好氣性微(wei)生物的小型試驗設備(bei)或作為種子擴(kuo)大(da)培養(yang)之用,常用的搖(yao)(yao)床(chuang)有往復(fu)式和(he)旋(xuan)(xuan)轉(zhuan)式兩(liang)種。往復(fu)式搖(yao)(yao)床(chuang)的往復(fu)頻率(lv)一(yi)般在(zai)80~140次(ci)/min,沖(chong)程(cheng)一(yi)般為5~14cm,如頻率(lv)過(guo)(guo)快(kuai)、沖(chong)程(cheng)過(guo)(guo)大(da)或瓶內液(ye)體裝量過(guo)(guo)多,在(zai)搖(yao)(yao)動(dong)時液(ye)體會(hui)濺到(dao)包扎瓶口的紗布(bu)或棉塞上,導(dao)致雜菌污染,特別是啟動(dong)時更容易(yi)發生這種情(qing)況。旋(xuan)(xuan)轉(zhuan)式搖(yao)(yao)床(chuang)的偏(pian)心距一(yi)般在(zai)3~6cm之間,旋(xuan)(xuan)轉(zhuan)次(ci)數為60~300rpm。

放在(zai)搖(yao)(yao)床上的(de)培(pei)養瓶(ping)(ping)(ping)(一般(ban)為三角瓶(ping)(ping)(ping))中的(de)發酵液(ye)所(suo)需要的(de)氧(yang)是由空氣經瓶(ping)(ping)(ping)口包扎的(de)紗布(一般(ban)8層)或(huo)(huo)棉(mian)塞(sai)通入的(de),所(suo)以氧(yang)的(de)傳(chuan)遞與瓶(ping)(ping)(ping)口的(de)大小、瓶(ping)(ping)(ping)口的(de)幾何形狀(zhuang)、棉(mian)塞(sai)或(huo)(huo)紗布的(de)厚度和(he)密度有關。在(zai)通常情(qing)況下,搖(yao)(yao)瓶(ping)(ping)(ping)的(de)氧(yang)吸收系(xi)數(shu)取決(jue)于搖(yao)(yao)床的(de)特性和(he)三角瓶(ping)(ping)(ping)的(de)裝樣量。

往返式搖床是借助曲柄方法帶領搖床作往返運轉,屏幕為鐵或廚衛電器的長方框子,有顆層至三種拖盤,拖盤上都有園孔備放教育養育瓶,孔中凸起一名角形形橡皮擦,什么的工具固定不動教育養育瓶并抑制瓶的噪聲,齒輪傳動公司通常情況下適用特殊三角皮帶款的輪加速運動,置換改善三角皮帶款的輪可變動往返概率。剪力輪上打有各種不同的剪力孔,便于改善剪力距。往返式搖床的概率和剪力距的多少對氧的汲取有很明顯的干擾。 滑動式搖床是回收利用滑動的扁心軸使餐盤擺動旋轉,餐盤有條層或2層,能作不銹角鋼、鋁單板或竹木板手工制造。在3個扁心軸里裝有六角螺栓可調式節高低,使餐盤恢復技術。一些搖床組成繁瑣,造價比高。其優勢:是氧的傳承好一點、效率需求小、激發基不會輕易濺到瓶口的繃帶上。 顯微鏡觀察 微菌物生命戶細微,應該也是借助體視體視顯微鏡就可以考察了解清楚它的生命戶要素和血細胞構成。因而,在微菌物學的各種探究中,體視體視顯微鏡就將成為必須短缺的器具。

液體(ti)(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)(zhong)(zhong)接(jie)種(zhong)(zhong)(zhong)(zhong)(zhong)箱(xiang)是專(zhuan)為(wei)移(yi)接(jie)液體(ti)(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)(zhong)(zhong)而設(she)(she)計的(de)。比固體(ti)(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)(zhong)(zhong)箱(xiang)窄(zhai)長,單側兩(liang)人操作。內設(she)(she)軌道和紫外線燈,箱(xiang)兩(liang)端開有(you)(you)高25cm,寬10cm的(de)長方(fang)形出(chu)(chu)口(kou),方(fang)便菌(jun)(jun)種(zhong)(zhong)(zhong)(zhong)(zhong)進(jin)(jin)(jin)出(chu)(chu),洞(dong)口(kou)設(she)(she)有(you)(you)小(xiao)推(tui)門。進(jin)(jin)(jin)出(chu)(chu)口(kou)下處設(she)(she)蒸汽源,接(jie)種(zhong)(zhong)(zhong)(zhong)(zhong)時用蒸汽封住進(jin)(jin)(jin)出(chu)(chu)口(kou),以防雜菌(jun)(jun)進(jin)(jin)(jin)入箱(xiang)內。箱(xiang)背(bei)面設(she)(she)有(you)(you)液體(ti)(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)(zhong)(zhong)移(yi)接(jie)管(guan)能進(jin)(jin)(jin)入的(de)小(xiao)孔。

接(jie)(jie)種(zhong)箱滅(mie)菌時,用紫(zi)外(wai)(wai)線(xian)照射30min。如果沒有(you)紫(zi)外(wai)(wai)線(xian)燈,可用甲醛和高(gao)(gao)錳(meng)酸(suan)鉀(jia)(甲醛10~14mL/m3+高(gao)(gao)錳(meng)酸(suan)鉀(jia)5~7g/m3空間(jian))熏(xun)蒸30min以上。使用時,先將所需(xu)物品(pin)和工具放入接(jie)(jie)種(zhong)箱內,然后進(jin)(jin)行藥(yao)劑熏(xun)蒸和紫(zi)外(wai)(wai)線(xian)滅(mie)菌,再按無菌操作進(jin)(jin)行接(jie)(jie)種(zhong)。接(jie)(jie)種(zhong)箱的結構簡單,造價低(di)廉,易消(xiao)毒(du)滅(mie)菌,操作方便,而且人在(zai)箱外(wai)(wai)操作,氣(qi)溫較高(gao)(gao)時也能(neng)作業。缺點(dian)是進(jin)(jin)出培養基費工費時,每次接(jie)(jie)種(zhong)前都需(xu)要(yao)進(jin)(jin)行滅(mie)菌。

家用冰箱 微生物實驗室的冰箱主要有兩種:普通冰箱和低溫冷凍冰箱。普通冰箱一般都具有兩個柜子,即鮮藏柜和冷藏柜,溫度分別為4℃和-20℃;低溫冷凍冰箱溫度一般控制在-40~-80℃。它們都可以用于微生物菌種保藏。鮮藏柜常用于保存斜面菌種,保藏時間在3個月左右。超過3個月,斜面就會變干,因此需要轉接菌種。如果要長時間保存菌種,則需要經過處理后,貯藏于普通冰箱的冷藏柜或低溫冷凍冰箱中,它們的保藏時間較長,一般都在1年以上。
